Also known as acid fast stain developed to identify acid fast bacteria i.e the bacteria should be able to resist decolouristion with acids.
METHOD:
Basketball A smear is taken and stained by a strong solution of fuchsin and then decolorised with 20% sulphuric acid and then counter stained with dye such as methylene blue,acid fast bacteria retain fuchsin stain whereas others take up the counter stain.Specially used to identify tuberculous bacilli due to presence of high lipid, alchohols and fatty acid in the membrane.
